Morten Olsen's side dominated the first half and took the lead just after the half-hour mark when Daniel Agger slotted home from the penalty spot.
Australia tried to fight back after the interval but squandered a number of chances, with Stephan Andersen performing well in the Denmark goal.
Andreas Bjelland then scored in the 66th minute to double Denmark's advantage and put the result beyond doubt.
Denmark face a tough task to advance to the quarter-finals in Poland and Ukraine after being pitted in a group containing Germany, Portugal and Holland.
