The American had blown a big lead in the same event two years ago before losing in a play-off and once again found himself leading the way after picking up three shots on the front nine.
This time he held his nerve and although he closed with successive bogeys, nearest challenger and playing partner Jim Furyk also struggling home to take the pressure off the eventual champion.
"It probably has not sunk in," Dufner said. "I can't believe this is happening to me. To come back from a couple of years ago when I lost in a play-off feels really really good.
"I decided that I was going to be confident and put my best foot forward and play aggressive to try to win this thing.
"The last two holes were a little unfortunate but I am happy to get the job done and it's a big step in my career."
